Vacquinol-1 dihydrochloride is potentially used as a therapeutic for Glioblastoma multiforme (GBM). This drug is highly stable in vivo, with a half-life in plasma of 52 hour. Vacquinol-1 has high oral bioavailability and good brain penetration.
Vacquinol-1 (NSC13316) is a brain penetrant inducer of catastrophic vacuolization in glioblastoma cells (GCs). Vacquinol-1 displays high cytotoxicity to GCs; it induces cell death by membrane ruffling, cell rounding, massive macropinocytic vacuole accumulation, ATP depletion, and cytoplasmic membrane rupture of GCs. Vacquinol-1 originally was developed as an antimalarial compound.
